Gene found for rare disease that rapidly ages kids

WASHINGTON (Reuters Health) - Scientists from the U.S. and France simultaneously announced on Wednesday the discovery of a gene that causes progeria, an extremely rare disease that prematurely ages and kills children.

The finding was hailed by researchers as extraordinary, because it took just under a year to pinpoint the gene.

And, since the gene causes premature aging, it could play a role in normal aging and in contributing to heart disease and stroke, said Francis Collins, director of the National Human Genome Research Institute, and a contributor to the progeria gene hunt.

"What we learn about the molecular basis of this model of premature aging may provide us with a better understanding of what occurs in the body as we grow older," Collins said at a press briefing.

Progeria has been hard to study since it occurs only one or two times in every eight million births worldwide, and only a few dozen children are alive with the condition at any given time, said Ted Brown, another progeria gene hunter and a geneticist at the New York State Institute for Basic Research in Developmental Disabilities. About one baby is born with the condition per year in the U.S.

Also, progeria is not an inherited condition, which has puzzled many scientists over the 110 years since it was first noted.

Children with the condition fail to thrive, leading to very short stature, loss of body fat and joint stiffness. By the time they are four or five years old, they look like wizened old people. With the premature aging comes heart disease and hardening and clogging of the arteries, making stroke and heart attack the leading cause of death in the children, who rarely live beyond adolescence.

The hunt for a genetic cause took off quickly after Leslie Gordon, a physician whose son was born with the disease, began a crusade in 1999 to fund more research. She convinced the National Institutes of Health to hold a workshop on progeria in 2001.

The researchers had some clues -- past studies of progeria children seemed to indicate an abnormality in the region of chromosome one. Their hunt was helped along by the continuing completion of the map of the human genome.

By October 2002, a consortium of NIH scientists and researchers from around the country had narrowed their search to a single gene, the lamin-A. Normally, that gene produces proteins that help the cell's nucleus hold all its contents during cell division.

In 18 of 20 children with progeria studied, U.S. researchers found an error in the body's copying of genetic material from one cell to the next. There was a misspelling in one single DNA base among 25,000 base pairs. The error, located on chromosome one, led to a shortening of a protein produced by lamin-A.

Children with progeria had very misshapen cell nuclei, which likely leads to widespread cell death and premature aging, concluded lead author Maria Eriksson of the NHGRI in a paper announcing the gene discovery published this week in the journal Nature.

The French researchers reported the same findings in the journal Science.

Gordon of the Progeria Foundation said that the work "is a great springboard" from which to find diagnostic tests, treatments, and perhaps, one day, a cure for the condition.

Tests are just around the corner, said Collins.

Nog even oefenen op je engels Haha!

Progeria is een ziekte waardoor kinderen heel snel verouderen en uiteindelijk sterven van "ouderdom". Ze zijn dus nog heel jong, maar hebben het lichaam van een bejaarde.

Tuurlijk!

Het komt er op neer dat wetenschappers in minder dan een jaar tijd (uitzonderlijk!) het gen dat progeria veroorzaakt gevonden hebben.

Dit gen zou ook een rol kunnen spelen bij de normale veroudering van mensen waardoor bijvoorbeeld hartkwalen ontstaan. Als ze er achter komen hoe die snelle veroudering in z´n werk gaat, dan komen ze waarschijnlijk ook meer te weten over normale veroudering.

Progeria is moeilijk te onderzoeken omdat het heel zeldzaam is. Maar 1 of 2 op de 8 miljoen geboortes. Ook is het geen erfelijke aandoening.

Wat progeria is weet je he? Sla ik dat stukje even over Haha!

Dan komt er een stukje over hoe en wie dat onderzoek gedaan hebben en ook nog wat wetenschappelijke details over chromosomen etc. Voert een beetje te ver om dat te vertalen en ook nog eens uit te leggen.

Dat het gen gevonden is, is een basis voor diagnostiek, tests, behandeling en misschien uiteindelijk genezing. Het zal iig niet lang meer duren voordat ze het 'makkelijk' kunnen testen.

Mmm, kan het bij beide niet vinden... In de Metro van vandaag als pdf staat het iig niet en op de website van de spits volgens mij ook niet.
